Title: Characteristics of immune gene rearrangements in Chinese Multiple Myeloma patients

Research Summary:
This study utilized NGS technology to systematically analyze the immunoglobulin (Ig) and T-cell receptor (TCR) gene rearrangement characteristics of 79 Chinese patients with multiple myeloma (MM). It was found that the proportion of Ig clones in the initial samples correlated with treatment efficacy. More importantly, the diversity of TCR in peripheral blood was significantly lower in patients than in healthy individuals. Based on this finding, a novel prognostic prediction model with extremely high discrimination (AUC=0.988) was developed, providing a new tool for liquid biopsy and prognostic evaluation in MM.

This study addresses the issue of CAR-T cell exhaustion in PD-L1-high large B-cell lymphoma by proposing a novel combinatorial therapeutic strategy: the use of 'armored' CAR-T cells expressing IL-7/CCL19 in conjunction with timed PD-1 blockade. Experimental results demonstrate that this combination effectively reverses CAR-T cell exhaustion, significantly enhancing their proliferation, persistence, and anti-tumor functionality, offering a new paradigm for overcoming tumor immune-suppressive microenvironments.

Title: Targeted capture sequencing reveals driver mutations and makes it possible to construct an accurate prognostic model for multiple myeloma

Research Summary:
This study performed driver mutation analysis on 101 newly diagnosed multiple myeloma (MM) patients using targeted capture sequencing. Based on high-frequency mutation profiles such as KRAS, NRAS, and TP53, a precise individualized prognostic risk model was developed. This model demonstrates excellent performance in predicting high-risk patients and significantly outperforms traditional staging systems, providing crucial support for precision diagnosis and treatment of MM.

During the conference, our team also engaged in face-to-face professional exchanges with several top domestic scholars to jointly discuss the clinical implications and future directions of the research findings.
